Experimental overview of the research project (work packages and tasks).

 

Work package (WP) 1: Exploring the presence of mesenchymal stem cells (MSCs) and their association with glioblastoma (GBM) cell and GBM stem cells (GSC) therapeutic sensitivity in patient-derived organotypic multicellular spheroids (OMSs) and patient clinical data

Task 1.1: Detection of MSCs in OMSs

Task 1.2: Evaluation of cytotoxic effects of irradiation and TMZ treatment on GBM cells and GSCs in OMSs

Task 1.3: Correlation of the amount of MSCs in OMSs with cell responses to treatment and clinical data

Realization: in progress, collecting tumor tissue samples and establishing OMSs

WP 2: Evaluation of the effects of MSCs on GSC therapeutic sensitivity in co-cultures in vitro

Task 2.1: Evaluation of GSC proliferation, apoptosis and cell cycle progression

Task 2.2: Evaluation of GSC self-renewal, stemness and differentiation

Task 2.3: Evaluation of DNA damage responses in GSCs

Task 2.4: Evaluation of GSC migration and invasion

Task 2.5: Analysis of cytokine release in co-culture media

Task 2.6: Evaluation of inhibition of candidate cytokine signaling on GSC treatment sensitivity in vitro

Realization: in progress, evaluating MSC effects on GSC proliferation, cell cycle progression, invasion and DNA damage response after treatments, searching for candidate cytokines

WP 3: Studying the effects of MSCs on GSC therapeutic sensitivity in zebrafish embryo brain in vivo

Task 3.1: Evaluation of cell proliferation and invasion

Task 3.2: Evaluation of GSC stemness, differentiation and expression of GSC niche proteins in mixed tumors

Task 3.3: Evaluation of inhibition of candidate cytokine signaling on GSC treatment sensitivity in vivo

Realization: in progress, evaluating MSC effects on GSC proliferation and invasion, optimizing preparation of tissue sections and immunohistochemistry for tumors for stemness and differentiation evaluation
