Their selectivity profile will be finely tuned by exploring several key parameters including i) binding affinity and selectivity of the recognition motif directed toward MMP catalytic cleft, ii) reactivity of the electrophilic warhead used to target Lysine, and iii) subtle orientation of this reactive motif within the S<sub>3<\/sub>\u2019 subsite of MMP.<\/li>\n\n\n\n<li><strong>Test the effect of MMP-2\/9 inhibitors<\/strong> in combination with irradiation in <em>in vitro<\/em> 3D GBM invasion models, comprising also tumour microenvironment.<\/li>\n<\/ul>\n\n\n\n<p>Project will be implemented in two work packages.<\/p>\n\n\n\n<p><strong>WP1:<\/strong> Design, screening and selection of covalent inhibitors targeting specifically MMP-2 and MMP-9 (MMP-2\/9), partner CEA<\/p>\n\n\n\n<p>  <strong><em>Task 1<\/em><\/strong><strong>.<\/strong> Design, synthesis and characterization of a focused library<\/p>\n\n\n\n<p>  <strong><em>Task 2<\/em><\/strong><em>.<\/em> In vitro evaluation of covalent inhibitors on a set of MMPs through enzyme assays enabling to determine their inhibitory\/selectivity properties<\/p>\n\n\n\n<p>  <strong><em>Task 3<\/em><\/strong>. For the best candidates, optimization of their reactivity in complex proteomes, consisting in modulating their structure to limit their unspecific reactivity toward abundant proteins<\/p>\n\n\n\n<p><strong>WP2: <\/strong>Test the effect of MMP2\/9 inhibitors and irradiation in <em>in vitro<\/em> GBM models of invasion and organoid invasion model, partner NIB<\/p>\n\n\n\n<p>  <strong><em>Task 4. <\/em><\/strong>Analyse protein levels and activity of MMP-2\/-9 in GBM cell model lysates<\/p>\n\n\n\n<p>  <strong><em>Task 5. <\/em><\/strong>Determine cytotoxicity of MMP2\/9 covalent inhibitors and irradiation in GBM cellular models<\/p>\n\n\n\n<p>  <strong><em>Task 6<\/em><\/strong><strong>.<\/strong> Perform GBM cell invasion assays with MMP2\/9 covalent inhibitors and irradiation<\/p>\n\n\n\n<p>  <strong><em>Task 7<\/em><\/strong><strong>.<\/strong> Perform organoid invasion assay<\/p>\n","protected":false},"excerpt":{"rendered":"<p>Project aims: Project will be implemented in two work packages.
